TRAP( tagged RNA affinity purification)是一種通過設計ncRNA-MS2載體與GST-MS2融合表達載體并共轉細胞,以檢測與ncRNA結合的蛋白質(RBPs)或RNA的研究方法。根據研究對象的不同,TRAP可與高通量測序、qPCR聯用,鑒定與ncRNA結合的RNA種類與相互作用水平;或是與MS、Western blot聯用,鑒定與ncRNA結合的蛋白質種類與結合強度。 (本試劑盒僅供科研用途)
實驗原理:
TRAP方法首先設計GST-MS2融合表達載體和ncRNA-MS2莖環結構串聯重復載體,共轉細胞獲得GST-MS2融合蛋白和ncRNA-MS2融合RNA,在胞內形成GST-MS2~ncRNA-MS2與蛋白或RNA的復合體;然后裂解細胞,利用谷胱甘肽親和瓊脂糖磁珠拉取獲得目的蛋白-RNA復合物;最后進一步分離純化,獲得目的蛋白或目的RNA進行下一步檢測分析。

3. 使用環狀RNA表達載體而非探針進行雜交,避免了circRNA研究中接頭序列被蛋白結合后探針不能結合circRNA導致的假陰性影響,且降低了探針雜交過程中RNA的降解。
